The HPLC method of identification of polyphenols in plant extracts by example of determination of isoflavone composition in soy seeds.

The method of identification of polyphenols in plant extracts for HPLC-analysis is proposed. The method is based on determining the degree of similarity of the identification characteristics of test substances and polyphenolic standards without the use the photodiode array detector. The identification characteristics include the retention time and the spectral characteristics. The heights of peaks of substance on the chromatograms at 255, 286 and 350 nm relatively the height at 225 nm are proposed to use as the spectral characteristics. The formulas for calculating the similarity indices of identification characteristics of the test substance and the standard and the criterion of their similarity are proposed. The proposed identification characteristic’s set improves the accuracy of identification of polyphenols. The method allows identifying the substance in the case of its compliance with several standards by retention time. The method allows identifying the different derivative forms of the same polyphenol (aglycons, glycosides) under the condition of similarity of their spectral characteristics. The procedure of researching of isoflavone composition in soy seeds of cultivar Farvater (Ukrainian breeding) are presented as an approbation of the proposed method. Isoflavone aglycons daidzein, glycitein, genistein, and their derivates 7-O-β-glucosides and 6’-O-malonylglucosides were identified in soy seeds.
